One unit is defined as the amount of enzyme required to digest 1 µg of lambda DNA in 1 hour at 37 °C in 50 µL of assay buffer.
Incubation of single stranded and double stranded radiolabeled oligonucleotides with 10 units of the enzyme for 4 hours at 37 °C.
After 160-fold overdigestion (10 U/µg Lambda DNA for 16 hours at 37 °C) with enzyme the fragmentation pattern is analysed. No detectable changes compared to the theoretical fragmentation pattern are considered as absence of star activity.
BglII is an endonuclease enzyme isolated from strains of Bacillus globigii lacking BglII and is part of the restriction modification system. In molecular biology it is used as a restriction enzyme that creates sticky ends. The nucleic acid sequence where the enzyme cuts is AGATCT, which as the complementary sequence is TCTAGA.
