pUC19 vector is also a circular double stranded DNA and is 2686 base pairs long. It is similar to pUC18, but possesses different restriction sites. This plasmid possesses
i) ampicillin resistance gene,
ii) the origin of replication derived from pBR322
and iii) the lac Z gene derived from E. coli.
Within the lac region is also found a polylinker sequence having unique restriction sites. It is a popular teaching aide for demonstrating plasmid DNA isolation, restriction digestion and cloning.
